Protective Effect of Probucol against Adriamycin-Induced Apoptosis in Cultured Rat Cardiac Myocytes

摘要: Purpose : In adriamycin(ADR)-induced cardiomyopathy, several different mechanisms are sug- gested. However, little information is available regarding the role of apoptosis. present study, we examined induction apoptosis on ADR treatment and anti-apoptotic effects probucol, a lipid-lowering drug, also studied changes bcl-2 expression in order to see molecular underlying effect probucol. Methods Cardiac myocytes were isolated from 3-day-old rats, cultured low(1μM) or high doses(10μM) for 24 hours. Probucol(50μM) was added 30 minutes before administration. Apoptosis determined by TUNEL staining, esti- mated immunocytochemistry. Results The number TUNEL-positive cells significantly increased both groups treated with ADR. probucol evident only low dose. addition, low-dose group its inhibited pretreatment Conclusion These results suggest that might play an important ADR-induced cardiotoxicity, partially prevented And not related depression bcl-2. Additionally, inhibition gene suggests another cell survival mechanism could be implicated action (J Korean Pediatr Soc 2000;43:746-754)
