摘要: The gene identification and kinetic characterization of (E)-2-(acetamidomethylene)succinate (E-2AMS) hydrolase has recently been described. This enzyme catalyzes the final reaction in degradation vitamin B(6) produces succinic semialdehyde, acetate, ammonia, carbon dioxide from E-2AMS. structure E-2AMS was determined to 2.3 A using SAD phasing. is a member alpha/beta superfamily utilizes serine/histidine/aspartic acid catalytic triad. Mutation either nucleophilic serine or aspartate resulted inactive enzyme. an additional residue active site causes be unstable likely structurally important. also provides insight into mechanism hydrolysis identifies several potential catalytically important residues.
