The site of HIV-1 integration in the human genome determines basal transcriptional activity and response to Tat transactivation

摘要: Because of the heterogeneity chromatin, site integration human immunodeficiency virus (HIV) in genome could have dramatic effects on its transcriptional activity. We used an HIV-1-derived retroviral vector, which green fluorescent protein is under control HIV promoter, to generate by infection 34 Jurkat clonal cell lines each containing a single HIV-1 vector. In absence Tat, 75-fold difference expression level between highest and lowest expressing clones was observed. Basal promoter activity low 80% moderate high remaining 20% clones. found that differences levels are due not controlled DNA methylation or histone acetylation. Tat activated transcription clone, inverse correlation observed basal inducibility Tat. These observations demonstrate chromatin environment influences gene activates independently environment.
