Detection of homocysteine by conventional and microchip capillary electrophoresis/electrochemistry

摘要: A method based on capillary electrophoresis (CE) with electrochemical (EC) detection for the determination of both total homocysteine (tHcy) and protein-bound (pbHcy) in plasma is described. Both end-column off-column amperometric were investigated. Off-column resulted a more sensitive assay (Hcy). The limit was 500 nM using EC response linear over range 1-100 microM. Therefore, this appropriate quantification Hcy physiological concentration ranges found all disease states. Methodologies tHcy pbHcy human investigated optimized concentrations obtained from healthy individual determined to be 2.79+/-0.31 nuM (n = 4) 3.37+/-0.15 microM 3), respectively. methodology then transferred microchip CE-EC format reduced glutathione (GSH) detected. Future work will focus development ancillary methodologies identify other forms vivo.
