摘要: Naturally occurring infections of Vaccinia virus (VACV) have been recognized in Brazil during the past 10 years. Human Brazilian (BVV) typically occur as a zoonosis transferred from affected dairy cows to their handlers. Outbreaks caused notable economic losses rural community region. The origins BVV are unclear but previous analyses shown that at least two distinct clades exist. aim this study was develop rapid and inexpensive process for identification differentiation should facilitate epidemiological ecological investigations including improved diagnosis Orthopoxvirus infections. A SYBR green quantitative real-time polymerase chain reaction (PCR) targeting hemagglutinin gene developed identify different populations BVV, VACV vaccine strains used smallpox eradication campaign (Vaccinia Lister (VACV-LIS) New York City Board Health (VACV-NYCBH)), currently available vaccines (VACV-NYCBH DRYVAX VACV-NYCBH Acambis 2000). Three primer combinations (one amplify many orthopoxviruses all vaccinia viruses described so far; one differentiate (VACV-LIS, 2000); clades) were designed work same annealing temperature conditions. In addition, these methods able detect orthopoxvirus viral DNA lesion biopsy material without need extraction.
