摘要: The stress produced by the coupling of reactive oxygen species (ROS) and endoplasmic reticulum (ER) has been explored extensively, but little is known regarding their roles in early development mammalian embryos. Here, we demonstrated that vitro-produced (IVP) bovine embryos was governed cooperative action between ROS ER stress. Compared with tension 5% O2, 20% O2 significantly decreased blastocyst formation rate cell survival, which accompanied increases levels sXBP-1 transcript, an indicator. In addition, treatment glutathione (GSH), a scavenger, levels, resulted increased survival rates. Importantly, stress-associated transcripts were reduced GSH developing Consistent this observation, tauroursodeoxycholate (TUDCA), inhibitor, improved developmental rate, trophectoderm proportion, survival. Moreover, transcript markedly supplementation TUDCA, suggesting possible mechanism governing mutual regulation Interestingly, knockdown XBP-1 both elevation decrease antioxidant transcripts, ultimately vitro competence Based on these results, IVP highly dependent coupled response oxidative stresses. These results increase our understanding mechanism(s) embryonic may improve strategies for generation high competence.
