Spermatozoa protein alterations in infertile men with bilateral varicocele

摘要: Among infertile men, a diagnosis of unilateral varicocele is made in 90% cases and bilateral the remaining cases. However, there are reports under-diagnosis among men that its prevalence greater than 10%. In this prospective study, we aimed to examine differentially expressed proteins (DEP) extracted from spermatozoa cells patients with fertile donors. Subjects consisted 17 diagnosed 10 proven as healthy controls. Using LTQ-orbitrap elite hybrid mass spectrometry system, proteomic analysis was done on pooled samples 3 5 men. From these samples, 73 DEP were identified which 58 expressed, 7 unique group 8 control group. Majority DEPs observed be associated metabolic processes, stress responses, oxidoreductase activity, enzyme regulation, immune system processes. Seven involved sperm function such capacitation, motility, sperm-zona binding. Proteins TEKT3 TCP11 validated by Western blot may serve potential biomarkers for varicocele. have demonstrated first time presence distinct reproductive functions altered Functional profiling provides insight into mechanistic implications varicocele-associated male infertility.
