摘要: The non-structural protein 1 (NS1) of dengue virus is a useful target for diagnostics infection since the abundantly circulating in blood during acute phase disease. Prior work has established that secreted NS1 levels plasma correlates with viremia and hence can also be used to diagnose patients at risk developing hemorrhagic fever. Thus detection antigens may benefit an early rapid diagnosis due its long half life blood. Here we describe simple efficient method expression Escherichia coli, which could potentially develop monoclonal bispecific antibodies point care diagnostics. E. coli codon optimized synthetic full-length gene serotype (DEN-1) was successfully cloned expressed very high-level as inclusion bodies. affinity purified refolded recombinant (rNS1) yield 230-250 mg/L bacterial culture. rNS1 immunize mice hybridoma development. polyclonal antiserum from animals immunized this found specifically recognize rNS1, thus demonstrating immunogenic nature protein. sensitive serum diagnostic assay monitor outbreaks.
