摘要: Retroviral DNA integration involves a coordinated set of cutting and joining reactions. We find that the IN protein Moloney murine leukemia virus (MoMLV) is only viral required to accomplish these reactions in vitro. has site-specific nuclease activity cleaves 2 nucleotides from sequence present at 3' ends MoMLV made by reverse transcription. This reaction generates recessed are normal precursors for integration. also possesses joins staggered cut, protein, target DNA. Short duplex oligonucleotides, corresponding DNA, serve as substrate both cleavage reactions; there no special requirements acts The products detected direct physical assay.
