摘要: A cytological and molecular analysis was performed to assess the genetic uniformity true-to-type character of plants regenerated from 20 week-old embryogenic suspension cultures meadow fescue (Festuca pratensis Huds.), compared protoplastderived obtained same cell suspension. Cytological variation not observed in a representative sample directly suspensions protoplasts isolated therefrom. Similarly, no restriction fragment length polymorphisms (RFLPs) were detected mitochondrial, plastid nuclear genomes analyzed. Randomly amplified polymorphic DNA markers (RAPDs) have been used characterise molecularly set mature these vitro cultures. RAPD using 18 different short oligonucleotide primers arbitrary nucleotide sequence combination with polymerase chain reaction (PCR) allowed detection pre-existing donor genotypes, but failed reveal newly generated protoplast-derived their equivalent suspensionculture materials. The stability therefrom its implications on gene transfer technology for this species are discussed.
