TP53 mutations, amplification of P63 and expression of cell cycle proteins in squamous cell carcinoma of the oesophagus from a low incidence area in Western Europe

摘要: In Europe, high incidence rates of oesophageal squamous cell carcinoma (SCCE) are observed in western France (Normandy and Brittany) north-eastern Italy. Analysis TP53 mutations tumours from these regions has shown a prevalence at A:T basepairs that may result DNA damage caused by specific mutagens. However, the spectrum low is unknown. We report here mutation analysis 33 SCCE collected Lyon, an area incidence. These were also examined for MDM2 P63 amplification, expression p16INK4a/CDKN2a, cyclin E, p27Kipland Cox2. detected 36% cases (12/33). contrast with incidence, did not show base pairs. was amplified 5/32 tested (15.5%). No amplification found. Expression studies revealed frequent loss p16INK4a/CDKN2a(46%) p27Kipl(25%) expression, overexpression Cyclin E (70%) Cox2 (42%).
