摘要: Abstract Background & Aims: One-kilobase sequence of the upstream fragment pig lactase-phlorizin hydrolase gene has been shown to control small intestinal–specific expression and postweaning decline in transgenic mice. The aim this study was identify regulatory DNA elements transcription factors controlling expression. Methods: activity different promoter fragments investigated by transfection experiments using Caco-2 cells. Electrophoretic mobility shift assays supershift analyses were used characterize interaction between intestinal identified elements. Results: Functional analysis revealed three previously undescribed regions promoter: a putative enhancer −894 −798 binding hepatocyte nuclear factor (HNF)-1 at position −880; repressor-binding element −278 −264 which an HNF-3–like is able bind; −178 −164 that binds activating factor. Conclusions: Identification new HNF-1 as players regulation impact on understanding molecular mechanisms behind age-dependent, tissue-specific, differentiation-dependent, regional intestine.
