摘要: Abstract Objective: The objective of this study was a description changes in gene expression that occur response to mechanical injury cultured human saphenous vein. Methods: Restriction fragment differential display (Display Systems Biotech, Vista, Calif) used for the comparison profile seven sets vein, with first set representing at time harvest vein and other six different lengths culture or without crush injury. All were from single, freshly harvested Each contained eight separate segments. (Set 1) taken RNA immediately extracted. This reflects vivo harvest. Three segments (Sets 2, 4, 6) 24, 48, 72 hours after (culture only). 3, 5, 7) harvested, crush-injured, then (crush + culture). profiles these compared obtained Results: resulted up-regulated 21 identified (including inducible nitric oxide synthase) nine unknown genes down-regulated within had five platelet-derived growth factor–B transforming factor–β2) pattern regulation transmural revealed whose products support cell migration oppose proliferation. Conclusion: between those designated only shows some cells express phenotypic are consistent migration. Further studies vitro may elucidate endogenous vascular tissue injury. (J Vasc Surg 2002;35:146-51.)
