Flow cytometric analysis of G1‐ and G2/M‐phase subpopulations in mammalian cell nuclei using side scatter and DNA content measurements

摘要: Several subcompartments of the cell cycle in addition to G1-, S-, and G2-phases usually observed were identified by simultaneous flow cytometric measurements ethidium bromide fluorescence side scatter intensity nuclei. Metaphase cells very early G1-phase (G1A) with low intensities discriminated from interphase high intensities. The reason for various was found be different structure metaphase due chromatin condensation as shown sorting respective G1A-phase could further subdivided into two compartments (G1A1) intermediate (G1A2) Using partially synchronized duration these estimated. durations G1A1- G1A2-phases about 10 min 20 min, respectively, compared total 3 h. Additional nuclei provide therefore information on G1- G2/M-phases.
