摘要: Radiolabelled and fluorescent-dye-conjugated oligonucleotide probes which targeted rRNA sequences were developed for the enumeration of ruminal bacterium Synergistes jonesii 78-1 in mixed culture. Two tested, both highly specific respective complementary target organism. Individual cells S. pure cultures clearly visualized situ by hybridization with probe but could not be detected natural samples. Therefore radiolabelled was used to monitor population introduced into a chemostat simulated rumen ecosystem. The did hybridize RNA extracted from culture prior inoculation After inoculation, represented 4.5% total bacterial then rapidly declined < 0.2% before increasing about 1% during following 3 weeks. This study demonstrates that rRNA-targeted tracking organisms
