Promotion of EGF receptor signaling improves the quality of low developmental competence oocytes.

摘要: Abstract Oocytes acquire developmental competence with progressive folliculogenesis. Cumulus oocyte complexes (COCs) from small antral follicles have inherent low and are poorly responsive to amphiregulin (AREG) which normally mediates maturation ovulation. Using porcine COCs, in an vitro AREG-induced system, the combined exposure N(6),2ʹ-O-dibutyryladenosine 3′:5′ cyclic monophosphate (cAMP) bone morphogenetic protein 15 (B15) growth differentiation factor 9 (G9) was necessary enhance rate of meiotic blastocyst formation. Furthermore, combination cAMP+B15+G9 enabled AREG-stimulated cumulus expansion increased expression matrix-related genes HAS2 , TNFIPA6 PTGS2 . Additionally, enhanced p-ERK1/2 is downstream EGF receptor. The nuclear formation rates combinational treatment were ablated by receptor phosphorylation inhibitor. These results indicate that cAMP oocyte-secreted factors cooperate promote functionality developing representing a key component acquisition competence.
