摘要: Five serological assays were evaluated for the diagnosis of brucellosis in goats: rose bengal test (RBT), complement fixation (CFT), radial immunodiffusion (RID) with Brucella and Yersinia enterocolitica O:9 polysaccharides, counterimmunoelectrophoresis (CIEP) cytosol, enzyme-linked immunosorbent assay (ELISA) polyclonal protein G conjugates smooth lipopolysaccharide (S-LPS), native hapten polysaccharide (NH), or cytosol antigens. For optimal sensitivity, RBT had to be used sera-antigen at a 3:1 dilution. In RID test, melitensis biotype 1 NH could not replaced by abortus Y. 0:9 polysaccharides. ELISA, S-LPS gave similar results conjugate increased specificity. With sera from 55 B. culture-positive goats, sensitivity was 100% RBT, CFT (titer > = 4), ELISA NH; 94% RID; 93% CIEP. All tests negative (100% specific) when testing 127 brucella-free goats. Larger discrepancies among obtained goats areas where is endemic. When 20 young vaccinated subcutaneously (10(9) CFU Rev 1) bled 6 months later examined, specificities as follows: 60%; 75%; 80%; CIEP, 90%; RID, 94%. 10 conjunctivally all specific 4 after vaccination. The proportion giving positive reaction vaccination decreased faster than other tests.
