Cellular imaging and mitochondria targeted photo-cytotoxicity in visible light by singlet oxygen using a BODIPY - appended oxovanadium(IV) DNA crosslinking agent

摘要: Oxovanadium(IV) complexes VO(L-1)Cl-2] (1) and VO(L-2)Cl-2] (2), where L-1 is N,N,N-donor benzyldipicolylamine L-2 the BODIPY-appended base, viz., (8-{bis(2-pyridylmethyl)amino]methylphenyl}-4,4-difluoro-1,3,5,7-te tramethyl-4-bora-3a,4a-diaza-s-indacene), were synthesized, characterized their photo-induced DNA crosslinking activity photocytotoxicity studied. Single crystal X-ray diffraction analysis of 1 reveals two chloride ligands in cis disposition to VO2+ moiety. The are : electrolytes aqueous media, loss second takes place on irradiation with visible light. melting alkaline agarose gel electrophoresis studies complex 2, treated ct-DNA light, suggest formation crosslinks. Complex 2 selectively localizes mitochondria HeLa MCF-7 cancer cells shows remarkable via an apoptotic pathway (IC50 < 3 mu M) light (400-700 nm) low dark toxicity. showing dual activity: (i) light-activated VO2+-DNA crosslink (ii) singlet oxygen (O-1(2)) induced mitochondria-targeted PDT involving BODIPY
