摘要: Application-specific validation of antibodies is a critical prerequisite for their successful use. Here we introduce an automated approach characterization and screening against synaptic molecules high-resolution immunofluorescence array tomography. The method, based on probabilistic synapse detection algorithm, designed to provide robust, flexible, efficient tool antibody characterization. By allowing the user define molecular composition size synapses expected contain antigen, method applicable with different distribution. output algorithm are automatically computed characteristics such as density target specificity ratio, which reflect sensitivity immunolabeling given antibody. These measurements objective way characterize compare performance same target, can be used objectively select best suited AT, potentially also other applications.
