Evidence that APP gene copy number changes reflect recombinant vector contamination [preprint]

摘要: Abstract Lee et al.1 (hereafter “the study”) have recently reported that RNA-mediated somatic recombination or retrotransposition of the APP gene occurs in neurons from both control individuals and those with sporadic Alzheimer’s disease (AD). As evidence retrotransposition, authors present various forms genomic cDNA (gencDNA) PCR-based (Sanger sequencing, SMRT sequencing) non-PCR-based (targeted hybrid-capture DNA situ hybridization (DISH)) experiments. They also report greater prevalence gencDNA AD compared to (69% vs 25% at least one retrogene insertion on average, Fig. 5 Extended Data study) as well its diversity. We reanalyzed APP-targeted sequencing data study, revealing originates mainly contamination by exogenous recombinant vectors, rather true endogenous APP. our own single-cell whole genome (scWGS) show no for normal ages.
