摘要: High-performance liquid chromatography (HPLC) has been used to separate and identify the metabolites formed from thyrotrophin-releasing-hormone (TRH) its hyperactive analogue, (3Me-His)TRH, by subcellular fractions rat cortex, hypothalamus thalamus. Deamidation proline endopeptidase formation of histidylproline diketopiperazines pyroglutamyl aminopeptidase were found be major mechanisms brain inactivation both peptides; (3Me-His)TRH was slightly more stable than TRH in presence peptidases, with enhanced receptor binding affinity, this could explain increased biological activity. The HPLC system may applicable determining other analogues also define pathways for larger neuropeptides as well.
