Cationic channels in normal and dystrophic human myotubes

摘要: Human skeletal muscle cells obtained from normal and Duchenne muscular dystrophy patients were cocultured with explants of rat dorsal root ganglions. Single-channel recordings performed the cell-attached configuration patch-clamp technique negative pressure was applied via patch-pipette in order to mechanically stimulate membrane patch. Inward elementary current activity recorded under control or conditions. Its occurrence mean open probability higher dystrophy. Amplitude histograms reveal that these channels have a small unitary conductance around 10 pS 110 mM Ca2+ could be inhibited dose-dependent manner by gadolinium. Results show stress favoured calcium permeation through channels. Taken together data provide arguments for involvement such overload previously observed dystrophic human (Duchenne dystrophy) cells.
