摘要: Background: In late 2011, a new Orthobunyavirus of the Simbu serogroup named Schmallenberg virus (SBV) emerged in continental Europe. The is transmitted by hematophagous arthropods, with Culicoides species as, so far known, main vectors. Infection can cause clinical signs adult ruminants including diarrhea, fever and reduced milk production. Transplacental infection developing fetus lead to malformations varying severity. To assess seroprevalence SBV Sweden an indirect enzyme-linked immunosorbent assay (ELISA) was established connection surveys. Here, we describe development evaluation ELISA, based on whole as coating antigen monoclonal antibody for detection antibodies ruminant sera. includes comparison between in-house neutralization test commercial ELISA. Results: optimal working dilutions antigens conjugate were estimated checkerboard titrations. Comparative studies, ROC analyses, used selection cut-off (S/P value=sample value percentage positive control value). With S/P 15% ELISA showed specificity 100% sensitivity 99.19% compared (VNT) good consistency shown reproducibility variability experiments. Furthermore, our nucleoprotein antigen, demonstrated higher test. Conclusion: described this paper readily available serological analysis antibodies. Since demonstrates comparable also shows commercially it useful alternative surveillance screening purposes SBV.
