Localization of the hair cell's transduction channels at the hair bundle's top by iontophoretic application of a channel blocker.

摘要: In order to understand how the hair cell's mechanoelectrical transduction channels are gated during mechanical stimulation, it is essential determine their location with respect bundle's constituent stereocilia. We localized by focally blocking receptor currents iontophoretically ejected gentamicin, an aminoglycoside antibiotic that acts as a reversible channel blocker. The drug was most effective when directed at top of bundle, whereas application bottom or cuticular plate had little no effect. Computer simulations agreed experimental data only were hypothesized occur near top. These results confirm located stereociliary tips.
