摘要: Structured RNA elements that protect transcripts from 5′→3′ degradation are becoming useful research tools. Here we show exonuclease-resistant structures (xrRNAs) Flaviviruses can be used to heterologous messenger RNAs (mRNAs) in budding yeast. Installation of xrRNAs ahead a downstream internal ribosome entry site (IRES) leads the accumulation partially-degraded mRNAs substrates for cap-independent translation LacZ reporter, yielding 30-fold increase measured β-galactosidase activity. Additionally, by monitoring dual-luciferase reporters xrRNA sequences do not interfere with progression an elongating ribosome. Combined these data indicate creative ways stabilize potentially applications.
