摘要: Bacterial toxins are commonly detoxified by chemical treatment in order to use them human vaccines. We have used site-directed mutagenesis of toxin genes obtain bacteria that produce naturally nontoxic mutants bacterial toxins, such as pertussis (PT), cholera (CT) and Escherichia coli heat-labile enterotoxin (LT). Genetically PT showed a superior safety immunogenicity animal models, phase I II clinical trials, protective efficacy the early late stage III trial, proving definitive extensive way genetic detoxification can, should, replace treatment. The results obtained with genetically inactivated LT CT indicate can be not only vaccines for systemic immunization ones produced conventional technologies, but suggest these type molecules may prototype design construction innovative totally new design, mucosally delivered preventive therapeutic
