摘要: Summary 1. In anaesthetized, open-chest dogs, 2-nicotinamidoethyl nitrate (SG-75) administered intravenously (0.3–1 mg/kg) or intraduodenally (3 produced decreases in systemic blood pressure, coronary resistance, heart rate and an increase sinus outflow, but virtually no change myocardial oxygen consumption atrioventricular conduction. The effects of SG-75 emerged within a few minutes after dosing lasted over about 1 h. 2. isolated, blood-perfused sino-atrial node preparations the dog into artery decreased slightly at dose which doubled flow through artery. 3. did not impair conduction even doses increased to more than twice basal level. 4. papillary muscle anterior septal scarcely affected force contraction artery, although further large it transient decrease contraction. extremely ventricular fibrillation. 5. dogs cardiac input was kept constant (0.01–1 ascending aorta venous return without changing output. 6. 2-Nicotinamidoethyl alcohol, denitrated compound SG-75, had vasodilator action comparable those parent compound. 7. These results indicate be potential antianginal drug having cardiodepressant actions properties uncharacteristic nitrates.
