摘要: Publisher Summary This chapter describes the production of a highly sulfated derivative dextran beads, and its use to isolate human plasma coagulation factors. For beads chlorosulfonic acid are added dry pyridine. The Sephadex G-50 is then absorbed on nearly all liquid. Further, sodium hydroxide dissolved in water rapidly with mixing neutralize were collected sintered glass funnel washed. Purification factors II, IX, X involve: barium citrate adsorption, ammonium sulfate elution, chromatography. Fractions containing activities X, or IX pooled concentrated using PM-10 membranes. After 24-hr dialysis at 4°C against 100 volumes 0.6 M NaC1-0.01 phosphate (pH 7.0)- 0.001 benzamidine, factor pools filtered stored 1-ml portions –70°C.
