摘要: A method for the integration of linear DNA into Dictyostelium genome is described. Restriction enzyme-mediated integration, or REMI, involves transformation cells with a mixture plasmid DNA, linearized restriction enzyme, along enzyme that capable generating compatible cohesive ends in genome. The stimulates cognate sites chromosomes, usually as single-copy insertion event and little collateral damage to REMI has proven useful genetic screens placing molecular markers at particular points Over past 15 yr, been used identify hundreds interesting genes based on their mutant phenotypes.
