摘要: A problem associated with use of the polymerase chain reaction to amplify specific DNA fragments from formalin fixed, paraffin wax embedded tissues is not infrequent failure amplification. One possible reason for this could be presence inhibitor(s), which interfere activity reaction. It has been shown that such inhibitor(s) exist when amplifying human beta globin gene (which exists in genomic as a single copy gene) routine clinical samples. variety methods remove were investigated. The results indicate are removed by proteinase K digestion, followed purification phenol/chloroform, and centrifugation through Centricon-30 membrane (30,000 molecular weight cut off).
