摘要: Ideal conditions for platelet reactivity testing are critical optimal selection of a P2Y12 inhibitor. Data inconsistent regarding the impact high-fat meals on test assessment. Participants included 12 healthy subjects not taking antiplatelet drugs after 12-hour fast. After baseline assessment, were given 600 mg dose clopidogrel. Four hours later, maximum inhibition was tested in fasting state by light transmission aggregometry (LTA), VerifyNow P2Y12, vasodilator-stimulated phosphoprotein (VASP), and whole blood (WBA). Subjects then provided meal, function evaluated two later. Change measured aggregation LTA primary endpoint study. The Wilcoxon Rank Sum used to compare change between non-fasting conditions. Spearman rho (ρ) correlation coefficient evaluate association following meal. No significant occurred maximal transmission, as assessed with 5 μM ADP (p = 0.15) 20 μM (p = 0.07). There area under curve (p = 0.03) but (p = 0.18). Although there no assay (p = 0.16), correlated initial value (Spearman’s p = 0.008). VASP WBA varied minimally. meal did significantly alter assessment commonly tests. Greater intra-subject variability existed optically-dependent compared non-optically dependent NCT01307657 .
