An in vitro EPR study of the free-radical scavenging actions of the lazaroid antioxidants U-74500A and U-78517F

摘要: Oxygen-based free radicals have been shown to play a major role in the acute destruction of neurons following cerebral ischemia and may be involved chronic neurodegeneration seen Parkinson's disease, Alzheimer's other conditions characterized by progressive death central nervous system. Drugs belonging group antioxidant compounds, collectively known as lazaroids, strong neuroprotective effects experimental models ischemia. However, specific mechanisms which these drugs reduce harmful actions not established. Using electron paramagnetic resonance (EPR) spectroscopy with spin trapping, we investigated interaction U-74500A, first-generation lazaroid, U-78517F, second-generation two species oxygen-based aqueous solution stable nitrogen-based radical diphenylpicrylhydrazyl dimethyl sulfoxide. Superoxide were generated action xanthine oxidase on hypoxanthine. Hydroxyl Fenton reaction involving ferrous iron hydrogen peroxide. Both lazaroids EPR signal all three radicals, but differ potency relative selectivity. These observations are consistent being scavengers suggest that involve direct interactions several different radicals.
