摘要: Pyrrolysine is the 22nd genetically encoded amino acid. For many years, its biosynthesis has been primarily a matter for conjecture. Recently, pathway synthesis of pyrrolysine from two molecules lysine was outlined in which radical SAM enzyme acts as mutase to generate methylated ornithine lysine, then ligated form an amide with ɛ-amine second lysine. Oxidation isopeptide gives rise pyrrolysine. Mechanisms have proposed both and ligase, structures now exist each, setting stage more detailed understanding how synthesized functions bacteria archaea.
