摘要: We have developed a two-site ELISA for measurement in serum of bone-related degradation products derived from C-terminal telopeptides type I collagen. The assay is based on the application two highly specific monoclonal antibodies against amino acid sequence AHD-beta-GGR, where aspartic residue (D) beta-isomerized. In one-step incubation procedure, containing cross-linked diisomerized EKAHD-beta-GGR peptides are captured by biotinylated antibody and peroxidase-conjugated antibody. generated complex then bound to streptavidin surface via biotin conjugate. Desalted urinary antigens used standardization, parallelism observed with samples. Results obtained <2.5 h, both inter- intraassay imprecision <8%. CrossLaps concentration was 1748+/-740 pmol/L (mean +/- SD) premenopausal women (n = 65) 2952+/-1325 group healthy postmenopausal 169). Serum One Step capable detecting significant (P <0.001) effect hormone replacement therapy retrospective study involving 22 women.
