Analysis of the Phthiocerol Dimycocerosate Locus ofMycobacterium tuberculosis EVIDENCE THAT THIS LIPID IS INVOLVED IN THE CELL WALL PERMEABILITY BARRIER

摘要: Among the few characterized genes that have products involved in pathogenicity of Mycobacterium tuberculosis, etiological agent are those phthiocerol dimycocerosate (DIM) locus. Genes biosynthesis these compounds grouped on a 50-kilobase fragment chromosome containing 13 genes. Analysis mRNA produced from this wild type strain showed region is subdivided into three transcriptional units. Biochemical characterization five mutants with transposon insertions demonstrated (i) complete DIM molecules synthesized cytoplasm M. tuberculosis before being translocated cell wall; (ii) genesfadD26 and fadD28 directly their biosynthesis; (iii) both drrC andmmpL7 necessary for proper localization DIMs. Insertional unable to synthesize or translocate DIMs exhibit higher wall permeability more sensitive detergent than strain, indicating first time that, addition important virulence factors, extractable lipids play role envelope architecture permeability. This function may represent one molecular mechanisms by which ofM. tuberculosis.
