摘要: Cloned segments of the mouse glycerol-3-phosphate dehydrogenase (GPDH) gene, Gdc-1, were used to screen a human library. Human clones obtained spanned 25 kilobases genomic DNA containing GPDH GPD1. The 4 kb sequence from 5'-flanking region and first exon GPD1 was compared with corresponding sequence. Both sequences share HindIII site located in what has proven be highly conserved 3' untranslated an upstream gene unknown function, D15Kzl. 3.6-kilobase segment between D15Kzl Gdc-1 provisionally termed promoter. Alignment promoter revealed two domains. An distal is approximately 900 base pairs length. A downstream or proximal consists 300 immediately TATA-like box contains fat-specific elements 1 2. Analysis chromatin structure four DNase I-hypersensitive sites. They present liver brown fat, which expression high, but absent spleen, low. Methylation studies showed it heavily methylated sperm. However, each adult somatic tissue had unique distribution nonmethylated sites could easily identified by its methylation pattern. These data suggest structural model that explains how differentially regulated many types cells.
