Glucose cryoprotectant affects glutathione-responsive antitumor drug release from polysaccharide nanoparticles

摘要: Abstract The aim of this work was to prepare polysaccharide-based nanoparticles (NPs) sensitive glutathione (GSH), and elucidate the effect concentration glucose used as cryoprotectant during freeze-drying on GSH-responsiveness. NPs were obtained via ionic interaction between negatively charged polysaccharides, chondroitin sulfate dermatan sulfate, positively thiolated chitosan (CSSH), crosslinking CSSH before or after formation with a disulfide-bond containing crosslinker, N,N′-bis(acryloyl)cystamine (BAC). freeze-dried at two different concentrations (0.5 5.0%w/w) then characterized methotrexate delivery systems, studying GSH drug release, efficacy against tumor cells cellular internalization. Non-loaded highly compatible murine fibroblasts showed suitable size for being in anticancer therapy. When methotrexate-loaded highest concentration, they lost their responsiveness vitro. Drug-loaded shown inhibit growth (HeLa CHO-K1) greater efficiency than free methotrexate, disregarding freeze-drying. Nevertheless, confocal microscopy studies revealed that internalization 5.0% is more difficult lower concentration. Thus, should be taken into account development intended release function levels, due specific interactions GSH.
