摘要: Mitotic spindles isolated from the diatom Stephanopyxis turris become thiophosphorylated in presence of ATP gamma S at specific locations within mitotic apparatus, resulting a stimulation ATP-dependent spindle elongation vitro. Here, using indirect immunofluorescence, we compare staining pattern an antibody against proteins to that MPM-2, mitosis-specific phosphoproteins, spindles. Both antibodies label poles, kinetochores, and midzone. Neither exhibits reduced labeling salt-extracted spindles, although prior salt extraction inhibits thiophosphorylation S. Furthermore, both recognize 205 kd band on immunoblots extracts. Microtubule-organizing centers brightly with MPM-2 intact cells. These results show functional are phosphorylated vivo We discuss possible role cytoskeletal control function.
