MPP(+)-induced mitochondrial dysfunction is potentiated by dopamine

摘要: MPP(+), the major metabolite of Parkinsonism-inducing compound MPTP, responsible for destruction nigrostriatal pathway in primates and rodents, has been assayed isolated rat liver mitochondria presence physiological concentrations dopamine or analogous melanin-dopamine. 5 microM MPP(+) 70 melanin-dopamine, but not alone, decreased heat production oxygen consumption a mitochondrial suspension activated with succinate ADP. Both oxidized plus also reductive power measured MTT. Mitochondrial swelling was observed, associated an increase membrane potential, as synergistic effect between low dopamine. It is suggested that cytosolic dopamine, by itself via its autooxidation products, may play relevant role toxicity MPP(+). A failure regulation storage/release could aggravate damage trigger neurodegenerative process underlying MPTP Parkinson's disease.
