摘要: Incorporation by RNA polymerases of BrUTP into both plant root tissue and isolated nuclei as a method for localization the sites transcription has been used. In this paper pea was used, under conditions employed, nearly all incorporation occurs in nucleolus, thus must be catalysed polymerase I. Immunofluorescence confocal microscopy shows that pattern consisting many small foci distributed widely through dense fibrillar component nucleoli. Immunogold labelling using silver-enhanced Nanogold probe at electron microscopic level confirms approximately 200nm diameter. Simultaneous fluorescence situ hybridization with to external transcribed spacer (ETS) region pre-rRNA structures revealed immunofluorescence are very similar. A portion rDNA (18S) also good correlation within nucleolus. On other hand non-transcribed intergenic (NTS) little coincidence incorporation, double 18S NTS probes difference localization. These results suggest most correspond single genes.
