摘要: As the Global Polio Eradication Initiative progresses towards its conclusion inactivated poliovirus vaccine (IPV) is increasingly being used on a routine basis to ensure that any re-introduced viruses do not spread. However current administration of conventional IPV (cIPV) includes risk wild seed escaping from manufacturing facilities. To address this IPVs could instead be prepared Sabin live attenuated strains. (sIPV) type 2 has been found induce lower level antibodies than cIPV. The reason (s) for difference clear as little known about molecular mechanisms underpin formaldehyde inactivation process. investigate process and consequences, study analysed effect different aspects biology. serotype shows greatest differences between sIPV cIPV, range strains with varied antigenic biological properties have using alternative chemicals. viral antigenicity immunogenicity assessed methods pre-release control batches various novel techniques including biosensor-based technique immunisation-challenge experiments in transgenic mice. Both virus strain chemical affected potency preparations. functionality RNA ability bind undergo conformational changes necessary enter target cell investigated real-time RT-PCR FACS flow cytometry. Inactivation modified prevented virions undergoing changes. This research will contribute better understanding cIPV help develop new/modified protocols produce improved immunogenicity.
