摘要: We have cloned and sequenced cDNAs encoding the variable regions of light (VL) heavy (VH) chains monoclonal antibody KC4G3. VL belongs to group II resulted from a V kappa-J kappa 2 recombination. VH IIId arose V-D9-JH3 The frameworks are respectively 84% 83% identical corresponding human consensus frameworks. deduced amino acid sequence contains an asparagine-linked glycosylation site in framework 3 (N74 I75 S76). determined that large fraction indeed glycolysated. constructed IgG1, human/mouse chimeric (by inserting murine KC4G3 Fv-encoding into plasmids Fc domain) expressed it SP2/0-Ag14 mouse myeloma cells.
