Subcellular distribution of adriamycin in the liver and tumor of 3LL-bearing mice

摘要: The apparent subcellular distribution of adriamycin (ADM) was investigated in the liver and primary or metastatic tumor tissue C57BL/6 mice bearing i.m. 3LL. ADM measured by a fluorimetric assay various cell components, nuclei (N), mitochondria (MT), microsomes (M) soluble fraction cytoplasm (SF), either vitro at times incubation vivo after drug injection. In both experimental conditions more than 50% accumulated nuclei, whereas only proportionally low amount has recovered other fractions. However, progressive increase percentage stored M particularly MT noted tumor, reaching 3 starting on basis 24 hr treatment. elimination half-life consistently longer total liver, suggesting that have higher capacity to retain drug. Work is progress evaluate whether this accumulation these sites related specific affinity persistent binding, like covalent binding macromolecules, possibly accounting for mitochondrial injury usually observed treatment with ADM.
