摘要: The in vitro production of proinflammatory cytokines after stimulation with Actinobacillus pleuropneumoniae and the relation these vivo disease caused by A. were investigated. Within 24 h, (serotype 1) preparations, including killed bacteria, bacterial culture supernatant, lipopolysaccharide, extracts, porcine pulmonary alveolar macrophages (PAM) produced significant (P < 0.05) amounts tumor necrosis factor-alpha (TNF-alpha) interleukin-1 (IL-1) as measured bioassays. supernatants containing interleukin-8 from PAM preparations showed neutrophil chemotaxis, while alone did not. After infection pleuropneumoniae, mean levels TNF-alpha IL-1 serum, bioassays, elevated 37- to 27836-fold for 11- 5941-fold higher within 4 d post-infection, depending on treatments, remained up Day 7. Both also detected lungs bioassays immunocytochemistry. results indicated that both secreted surface components can stimulate produce mediators. Neutrophil chemoattractants rather than are major factor causing acute lung inflammation. elevation pigs occurred coincident onset clinical disease.
