摘要: BACKGROUND: Advances in the past two decades dendritic cell (DC) biology paved way to exploit them as a promising tool cancer immunotherapy. The prerequisite for DC vaccine preparations is large-scale vitro generations of homogeneous, mature, and functional DCs. Frequent improvements are being made existing production protocols achieve this goal. In our previous study we reported generation DCs from umbilical cord blood (UCB) CD34+ cells. Here report that method can be used efficient UCB mononuclear cells (MNCs) thus hematopoietic stem isolation step not essential. STUDY DESIGN AND METHODS: MNCs or isolated same (CB) samples were characterized morphology, phenotype, assays including antigen uptake, chemotaxis, mixed leukocyte reaction. Similarly generated fresh frozen CB units compared. RESULTS: morphologic, phenotypic, characterization various sets show they comparable nature irrespective starting population used. CONCLUSION: We conclude essential eliminated. More importantly, with equal efficiency units. Our culture will useful exploiting potential an additional source allogeneic clinical settings.
