摘要: Several studies have established the potential efficacy of humoral immunity, primarily mannan-specific antibodies, in host protection against major fungal pathogen Candida albicans. In this study, we analysed immune response induced by immunization with BSA-based conjugates bearing synthetic α-1,6-branched oligomannosides (pentamannosides (M5) or hexamannosides (M6)) mimicking antigenic sequences cell wall mannan. We ability antibodies prepared to recognize relevant determinants mannan polysaccharide structure and C. albicans yeast hyphal morphoforms. M6-BSA conjugate markedly higher levels IgG compared M5-BSA conjugate. contrast conjugate, immunoglobulin isotype class switch from IgM IgG, as revealed also ELISPOT analysis. Immunization-induced showed reactivity form cells. The reduced immunogenicity seems be related branching point location at terminal non-reducing end comparison oligomannoside non-terminal location. Candidacidal activity assay different capacity sera improve candidacidal polymorphonuclear leucocytes. Limited – BSA induce significantly enhancing leucocytes was presumably absence strong corresponding natural activate complement. study documented structure-dependent limited branched α-mannooligosides production potentially protective antibodies.
