In vitro exposure with β-hydroxy-β-methylbutyrate enhances chicken macrophage growth and function

摘要: Abstract β-Hydroxy-β-methylbutyrate (HMB), a leucine catabolite, has been shown to decrease broiler mortality. One possible target of HMB action may be the cells immune system. Macrophages from chicken macrophage cell line, MQ-NCSU, were exposed 0, 10, 20, 40, 80, and 100 μg per 5 × 104 in 96-well culture plate. After 24 h exposure, proliferation was quantitated by an MTT bioassay. In duplicate experiments, stimulated growth over control (p ≤ 0.05) at wide range doses. 20 80 μg supernatant fractions tested for presence nitrite. exposure (20 μg) increased nitrite production 44.1% controls (Experiment 1, p ≤ 0.035). To determine phagocytic potential macrophages after MQ-NCSU line Sephadex-G50®-elicited abdominal incubated with fluorescent latex beads (1 : 40, ratio) 1 h then analyzed flow cytometry. When 40 μg HMB, significantly higher (31.7%) than that (p ≤ 0.0006). Sephadex-elicited exhibited 14.4% phagocytosis when treated (p ≤ 0.0016). Fc-receptor expression elevated (p ≤ 0.0001). These data demonstrate induces as well enhances effector functions, such phagocytosis. The findings these studies imply can used dietary immunomodulator.
