Leonurine (SCM-198) improves cardiac recovery in rat during chronic infarction

摘要: Leonurine, an alkaloid typically found in Herba leonuri, is known to have both antioxidant and cardioprotective properties. In the present study, we investigated mechanism of leonurine vivo rat model chronic myocardial ischemia vitro H9c2 cardiac myocyte oxidative stress. Myocardial was induced by ligating left anterior descending coronary artery. Rats were divided into sham, ischemia+saline, ischemia+leonurine (15 mg/kg/day). Cardiac function recorded catheterization. Apoptosis-related factor vascular endothelial growth (VEGF), survivin, Bcl-2 Bax pro-survival signaling pathways Akt, hypoxia inducible (HIF)-1α measured Western blotting or RT-PCR. Our results showed significantly improved as evidenced decreased ventricle end-diastolic pressure increased +dP/dt. Interestingly, phosphorylation protein gene expression Bcl-2, but it reduced vivo. Meanwhile Akt a concentration-dependent manner stress vitro, which abolished phosphoinositide 3-kinase (PI3K) inhibitor, LY294002. Furthermore, not only HIF-1α also survivin VEGF. The study demonstrated, for first time that has potent anti-apoptotic effects after mediated activating PI3K/Akt pathway. Angiogenic mechanisms may be partially responsible such effect, needs studied further.
