摘要: BACKGROUND:Cells release extracellular membrane vesicles including microvesicles known as exosomes. Exosomes contain microRNAs (miRNAs) however the full range within colorectal cancer cell secreted exosomes is unknown. OBJECTIVE: To identify of exosome encapsulated miRNAs from 2 lines and to investigate engineering over-expressing miRNAs. METHODS: were isolated HCT-116 HT-29 lines. RNA was extracted microRNA array performed. Cells engineered express miR-379 (HCT-116-379) or a non-targeting control (HCT-116-NTC) functional effects determined. by cells transferred recipient impact examined. RESULTS: Microvesicles 40–100 nm in size visualised confirmed exosomal protein CD63. contained 409 miRNAs, 393, 338 common both Selected targets validated. HCT-116-379 showed decreased proliferation (12–15% decrease, p< 0.001) migration (32–86% compared controls. enriched for miR-379. Confocal microscopy transfer cells. CONCLUSIONS: Colorectal secrete large number decreases migration, can be engineered.
