摘要: Replication of the two template strands at eukaryotic cell DNA replication forks is a highly coordinated process that ensures accurate and efficient genome duplication. Biochemical studies, principally plasmid DNAs containing Simian Virus 40 origin replication, yeast genetic studies have uncovered fundamental mechanisms fork progression. At least different polymerases, single-stranded DNA-binding protein, clamp-loading complex, polymerase clamp combine to replicate DNA. Okazaki fragment synthesis involves polymerase-switching mechanism, maturation occurs by recruitment specific nucleases, helicase, ligase. The also coupled cell-cycle progression repair maintain integrity.
